        Bio-Evaluation of 99mTc-Labeled Homodimeric Chalcone Derivative as Amyloid-β-Targeting Probe

        Introduction

        A substantial proportion of current and past literature has focused on utilizing amyloid plaque formation as the focal point to understand the etiology and pathogenesis leading to Alzheimer’s Disease (AD) and similar psychoneurological disorders (1, 2). The amyloid cascade hypothesis suggests sequential proteolytic processing of amyloid precursor protein (APP) by BACE (β-secretase) and γ-secretase into various smaller fragments of varying peptide lengths, of which Aβ40 and deleterious Aβ42 are the most common forms causing the formation of senile plaques in the human brain (3, 4). The Aβ assemblies encompass monomers, oligomers, and fibrils with varying aggregation susceptibility, among which oligomers have been associated with synaptic toxicity and plaques that are linked to the pro-inflammatory effects (4, 5). These different p
