The amyloid cascade hypothesis suggests sequential proteolytic processing of amyloid precursor protein (APP) by BACE (β-secretase) and γ-secretase into various smaller fragments of varying peptide lengths, of which Aβ40 and deleterious Aβ42 are the most common forms causing the formation of senile plaques in the human brain (3, 4). The Aβ assemblies encompass monomers, oligomers, and fibrils with varying aggregation susceptibility, among which oligomers have been associated with synaptic toxicity and plaques that are linked to the pro-inflammatory effects (4, 5).
